Gradient cyclers: Annealing optimisation and block formats

Gradient cyclers are thermal cyclers with the additional function of applying a defined temperature gradient across the thermoblock. This allows different annealing temperatures to be tested simultaneously within a single PCR run, enabling the determination of the optimal binding temperature for a primer pair without repeated individual runs. This function is particularly valuable when establishing new PCR assays or working with new primers.

ETC821 Series

The ETC821 models from ESPEC are compact gradient cyclers for daily laboratory use. They are available in three block variants: ETC821 with a 96-well block (compatible with standard high-profile PCR plates, strips, and tubes, promotional price €2,940.00), ETC821 with a 384-well block (promotional price €3,080.00), and ETC821DG with a 2×48-well dual block for two independent programmes simultaneously (promotional price €3,850.00). All models are designed for common PCR consumables without the need for special tips.

SensoQuest Labcycler Gradient

The SensoQuest Labcycler Gradient offers a flexible solution through interchangeable blocks: 48-well, 96-well, 96-well aluminium (for faster heating/cooling rates), and 384-well—one base unit for all formats. The gradient function is available for all block types. Price on request.

Distinction from Real-time Thermocyclers

Gradient cyclers are designed for classic endpoint PCR, where the amplification product is analysed after the run via gel electrophoresis or other detection methods. For direct quantification of nucleic acids in real-time (qPCR), the eQ-series real-time thermocyclers are available.
